项目摘要

糖代謝異常の病態下の妊婦におけるメチル水銀の代謝・動態や母児移行については明らかになっておらず、リスク評価・管理についても行われていない。本研究は糖代謝異常の病態下におけるメチル水銀の母仔移行の解明に資する知見の取得を目的とする。これまで、雌の糖代謝異常モデルマウスの確立に関する検討として、・二社の普通食または高脂肪食 (High Fat Diet: HFD)、・二通りの給餌期間（12週間または16週間）の比較実験を行い、BL/6マウスを用いた雌のDiet-Induced Obesity (DIO) モデルを得た。糖代謝異常の評価は、OGTTにより行った。さらに本モデルを用いて交配・妊娠・出産条件に関する実験を行った。しかしながら妊娠・出産率が低く、育児放棄等もあったため、解析に必要な検体が得られなかった。そこで膣内インピーダンス測定による性周期（交配適期）の推定を行った上で交配を行い、さらに実験系の切り替え（新生仔移行の評価から胎仔移行の評価へ）に関する検討を行った。現在、確立された実験系を用いて母親マウスにメチル水銀を投与し、得られた母親及び胎仔の各組織中の水銀濃度を測定中である。
