项目摘要

申請者は、胸腺摘出マウス(ATx)のT細胞において輸送膜タンパクであるMDR1が高発現し、機能的にも生体外物質を細胞外に排出しているという予備的実験結果をもとに、「MDR1が有害物質を細胞外に排出することで、胸腺退縮後の末梢性T細胞の生存延長・恒常性増殖促進に寄与している」という仮説を立てた。しかし、Ｔ細胞の恒常性増殖に作用するIL-7やIL-15を添加したin vitro培養や生体マウスの胸腺摘出後において、MDR1 KOマウス由来T細胞の生存率低下は認められなかった。MDR1阻害剤のベラパミルをATxマウスに投与する実験も行ったが、全身性副作用のために投与群で著しい体重減少をきたし、正確な評価はできなかった。以上の結果は、胸腺退縮後の末梢性Ｔ細胞の生理的な状況におけるMDR1の役割を示唆するものではなかった。元来MDR1は、生体外物質や脂質などを排出することが主な機能であるため、薬剤投与時や脂質異常症などの状況及び非リンパ臓器内のT細胞生存が低下している可能性は残された。これを検証するために、MDR1 KO及び野生型マウスを胸腺摘出したあとに高脂肪食を4週間摂取させ、T細胞の生存率を解析した。しかし、2次リンパ組織である脾臓に加えて肺・肝臓・脂肪組織においても、MDR1 KOマウス由来T細胞の有意な生存率低下を示唆するデータは得られなかった。現在は、サイトカイン無添加の条件で生存率が有意に低下するメカニズム、抗がん剤等の薬剤を投与した場合のT細胞生存におけるMDR1の役割など、残された課題点について検証を行っている。令和2年以降、申請者の所属する研究室では新型コロナウイルスに対するＴ細胞応答における加齢の影響を解析する橋渡し研究が開始され、申請者も参加した。その検体を用いてヒトT細胞におけるMDR1の役割を探索することも試みている。
