本研究では、他者を慰める行動の基盤となる神経機序を解明し、さらに自閉症のような精神疾患ではどのような原因で共感の欠如が生じるのかについて新たな知見を提供することを目標としている。令和4年度は、精神疾患横断型モデルマウス（22q11.2欠失モデルマウス）を用いて他者への慰め様行動に変化が見られるか検証を行った。この解析によって、他者への毛づくろい行動の表出が慰めを受ける相手個体の遺伝子型によって変化する可能性を示唆する結果が得られた。また、光ファイバーケーブルや小型顕微鏡を取り付けたマウスでも、他者への毛づくろい行動が表出するような実験条件の検討を前年度に引き続き行った。そして、帯状皮質（Cingulate cortex）に含まれる神経細胞がマウスの慰め様行動にどのような役割を果たしているか検証するため、カルシウムイメージング法による他者への毛づくろい行動中における帯状皮質の神経活動の測定、およびDREADDやオプトジェネティクスを用いた神経活動の抑制による慰め様行動の変化についての検証を実施した。次年度では、上記の実験を継続して実施するほか、神経活動の操作実験では、帯状皮質の神経活動を活性化した場合の慰め様行動の変化についても観察を行う。さらに、当初の計画していた22q11.2欠失モデルマウスに加えて、精神疾患との関連が示唆されている遺伝子の変異マウスについても、本研究で確立した実験系を用いることで、慰め様行動に変化が見られるか検証を行う。
