1. 本研究では、臨床的に、転移性脳腫瘍をよく再現するマウスモデル作製の確立と、免疫チェックポイント分子であるCD276を発現するM2マクロファージを主体とする慢性炎症の関与を明らかにし、これら細胞・組織群に対する標的分子の治療効果を臨床に近い形で検証する実験系を立案した。使用するマウスモデルについて既に作成済みであるが、今後もさらに、本研究により最適化されたマウスモデル作成を目指している。2. すでに、先行研究において、転移性脳腫瘍症例の血中M2マクロファージでの免疫チェックポイント分子CD276の発現亢進を見出しているが、転移性腫瘍組織における免疫チェックポイント分子CD276発現状況についてはこれまでの報告は少なく、これらを検証するべく、予備研究として計画している。3. 臨床的に転移性脳腫瘍において、免疫チェックポイント分子CD276を発現するM2マクロファージを主体とする慢性炎症の関与を明らかにするため、脳転移組織サンプル採取を行なっている。4. 2022年度は、マウスモデルの最適化及び、これらより脳組織を採取し、フローサイトメトリー解析、トランスクリプトーム解析、網羅的サイトカイン解析を行い、さらに、マウス糞便および腸管内容物は腸内細菌叢解析に供する予定であったが、一時的に持続継続的な実験が困難であった。
