本研究は①末梢血単核球のアミノ酸構成と臨床所見の比較、②iPS細胞から分化誘導した血管内皮細胞の細胞機能評価を行うことを研究全体の予定としている．本年度は①を中心に検討を行った．健常な若年成人3名とリジン尿性蛋白不耐症（LPI）患者３名（女性24歳，男性18歳，女性27歳）より末梢血単核球細胞を採取した．LPI患者3名のSLC7A7バリアントは全例c.1228C>Tであった．専用スピッツで単離した末梢血単核球細胞を用いて，細胞内アミノ濃度を健常人及びLPI患者を測定した．標準化した細胞内アミノ酸濃度の比較では二塩基性アミノ酸のうちリジン，アルギニンがLPI患者由来末梢血単核球で高値であった．二塩基性アミノ酸以外のアミノ酸でもバリン，プロリン，アラニン，システイン，グリシン，セリン，ヒスチジン，イソロイシン，スレオニン，グルタミンが高値であった．非上皮細胞における細胞内アミノ酸濃度もLPIの病態を形成しているアミノ酸トランスポターのバリアントの影響を受けていることが明らかとなった．

This study was conducted to compare the acid composition of peripheral blood mononuclear cells with clinical findings, and to evaluate the cellular function of vascular endothelial cells after differentiation induction of iPS cells. This year's event was held in the center of the city. 3 healthy adults with urinary protein intolerance (LPI)(female 24 years old, male 18 years old, female 27 years old) were enrolled in this study. The intracellular concentrations of these proteins were measured in healthy individuals and LPI patients. Normalized comparison of intracellular acid concentrations in patients with LPI resulted in high values in peripheral blood mononuclear cells. A dibasic acid is an acid other than a basic acid. It is a high value acid. It is a high value acid. In non-epithelial cells, intracellular acid concentrations and LPI pathologies are affected by acid concentrations and LPI pathologies.
