2013年4月～6月初旬、調査申請を調査地域(桜井市・橿原市・明日香村・大淀町・御所市・葛城市・高取町・吉野町・東吉野村)の教育委員会に申請し許可を受け調査を実施した。5月中は、各市町村の郷土史等文献調査を行った。その結果、生息の確認が記述されている桜井市(初瀬川、栗原川、寺川)・橿原市(飛鳥川、米川)・大淀町(竜門川、吉野川)・御所市(葛城川、曽我川)・吉野町(吉野川)・東吉野村(高見川)を中心に、6月10日から9月30日の期間で、オオサンショウウオの分布調査を聞取り、カニカゴによるトラップでの捕獲(33か所、355回)、夜間(午後7時～10時)に河川内での捕獲作業(初瀬川3回、飛鳥川3回、寺川2回、竜門川3回)によって行った。更に、吉野町の吉野川では目撃場所(水深約2メートル)約500㎡をスク一バダイビングによる潜水調査を昼間に実施した。延べ潜水時間は、3時間であった。結果、捕獲はできなかったが吉野町(紀ノ川水系吉野川)・東吉野村(同高見川)・桜井市(大和川水系初瀬川、同寺川)での生息の可能性が明らかになった。また、今回は調査申請をしていなかったが十津川村(新宮川水系十津川)や天川村(同天ノ川、弥山川)での生息確認の情報もそれぞれの地元の方から頂いた。今後、調査の申請を行い、調査を進めたい。繁殖生態は、木津川水系で行う予定で県の調査に同行し生息場所の特定を試みた。7月初旬の調査では、外鰓を持つ幼生(全長15㎝)が見つかり調査地域で繁殖していることが明らかになったが、その後繁殖巣穴を発見できなかったため繁殖生態を観察することができなかった。これについては、今後も繁殖巣穴を特定し観察を続けていきたい。
