脳動脈瘤の遺伝子治療という観点からウサギ脳動脈瘤発生モデルを用い、脳動脈瘤作製処置後6ヶ月後に脳動脈瘤の発生を確認できた動物に関し種々遺伝子の導入を行い動脈瘤の縮小化が得られるかを検討することが目的である.Basic fibroblast growth factor, platelet derived growth factor遺伝子をハイドロゲル・ポリマーに包含させ,これを手術用顕微鏡下で脳動脈瘤壁に塗布し,遺伝子導入を図り、3,6ヶ月後に脳動脈を摘出し、mRNAレベルをreverse transcription-polymerase chain reaction(RT-PCR)で定量的に分析し、蛋白質レベルを免疫染色法で定性的に分析した。さらに脳動脈瘤の縮小が得られるかどうかを組織学的に検討した.対照として治療処置を受けない群と比較を行った.3ヶ月後の変化としては,mRNAレベルはBasic fibroblast growth factor, platelet derived groth-factorともに有意な発現が見られ,免疫染色法による蛋白質レベルの検討でも両蛋白の血管壁における染色性の増強が主に中膜外層〜外膜に観察された.内膜〜内膜下には染色性は認められなかった.この時点での脳動脈瘤のサイズは対照群に比し,治療群では径として75%への縮小がみられ,かつ脳動脈瘤の壁が160%への増大が観察された.従って,これらの遺伝子の導入は脳動脈瘤の治療に応用出来る可能性があると考えられた.
