项目摘要

本研究は、口蓋裂の発生のメカニズムを解明し、臨床治療に応用するために、胎生期マウスの口蓋閉鎖に関わる分子機構を解析することを目標にしている。具体的には、口蓋突起の遺伝子発現を癒合前、癒合中、癒合後の3者間で比較し、口蓋閉鎖に関わる遺伝子群を探索することである。平成20年度に実施した研究の成果については、1.口蓋閉鎖に関わる遺伝子のスクリーニング(1)口蓋突起が接近した段階(癒合前)、(2)口蓋突起が癒合している段階(癒合中)、(3)口蓋突起が癒合した段階(癒合後)で、突起先端部のRNAを抽出し、T7RNA amplification法にてRNAを増幅し、T7-SAGE法を試みたが、時間を要したため、マイクロアレイ法を用いた遺伝子解析を開始した。2.口蓋突起上皮に特異的に発現する遺伝子の発現量確認癒合中・癒合後に、Carcinoembryonic antigen-related cell adhesion molecule 1 (CEACAM1)の発現量の増加が確認され、定量的real-time RT-PCRを用いても同様に発現量の増加が確認された。3.口蓋突起癒合部に特異的に発現する遺伝子の発現分布確認抗CEACAM1抗体を入手し、免疫組織学的解析を行うと、口蓋癒合中から癒合後にかけて、CEACAM1が口蓋突起先端部に強く発現していることが確認された。4.器官培養を用いた機能阻害実験機能阻害抗体にて口蓋癒合が阻害され、CEACAM1の口蓋癒合への関与が示唆された。現在のところ、細胞接着因子CEACAM1の器官形成に対する関与は未だに報告されていない。今後、他の遺伝子も含めて、口蓋癒合に対する関与とメカニズムについても解析を行う予定である。

Organ culture uses functional inhibition, functional inhibition antibodies, and CEACAM1 for inhibition of flap healing. The relationship between CEACAM1 and organogenesis has been reported. In the future, the relationship between his heritage and the healing of the mouth flap will be determined in advance.
