Elucidation of Mechanisms of HPV Carcinogenesis in Cervical Cancer

人类子宫颈癌的致癌机制尚未阐明。近年来，由于在宫颈癌细胞中发现了人乳头瘤病毒（HPV）基因组，因此认为HPV是宫颈癌的一个致病因素，本研究利用分子生物学技术检测了宫颈癌细胞中HPV基因组的存在。(1)Southern杂交法检测79%的癌前病变（异型增生）中HPV基因组阳性。(2)HPV 16 DNA通过转染小鼠Balb/3 T3细胞转化小鼠Balb/3 T3细胞。(3)这些结果表明HPV可能启动了宫颈癌的发生，某些辅助因子促进了宫颈癌的恶性进展。

The carcinogenesis of human cervical cancer of the uterus is not elucidated. Recently, human papillomavirus (HPV) is considered to be a factor, since the genome of HPV is detected in the tumor cells.We investigated the existence of HPV genome in the tumor cells of the uterine cervix using molecular biological techniques.(1) HPV genome was detected in 79% of the precancerous lesion (dysplasia) by Southern blotting method.(2) A HPV 16 DNA transformed mouse Balb/3T3 cells by transfection of the DNA.(3) Some steroid hormones enhanced the malignant progression of the transformants.These results indicate that HPV seems to initiate the carcinogenesis of cervical cancer and some cofactors promote the malignancy.
