進行期肺癌患者の末梢血中には高頻度にある種の血栓誘発因子(Thrombosis-Inducing Activity,TIA)の出現がみとめられる。我々はこの因子の物理化学的性質を解明することを目的に抗体の作製を試みた。人肺癌由来細胞株(QG56)に対する抗血清がTIA活性を中和する事が解ったので、この細胞株を抗原として用いTIA活性を中和しうるモノクローナル抗体を作製した。この抗体を利用してELIZA法によるTIAの定量測定法を確立した。TIAは癌細胞株の細胞質内に存在し培養上清中に分泌される物質であることがわかった。Western-Blotting法を用いてTIAの分子量の測定を試みた。しかし、その理由は不明であるが抗体によって染色される線を同定することが出来なかった。そこで、HPLCを用いて上清をゲル瀘過し各フラクションのTIA量をELIZAにより測定した結果、TIAは分子量20-30万の部分に認められた。モノクローナル抗体をSepharose-4Bゲルに結合させAffinity Purificationにより上清中からTIAの精製を試みたが牛胎児血清中に含まれる蛋白の非特異的結合が多く精製は困難であった。そこで、現在癌細胞株を無血清倍地で増殖させるべく試みている。動物を用いた癌転移実験で、TIAが癌の転移を著明に亢進させることがわかった。この結果は、人肺癌患者においてTIA陽性例が陰性例に較べて優位に予後が悪い結果の裏付けになりうるものと思われる。現在、TIAによる転移誘導の機構について検討中である。

Several thrombotic factors (Thrombosis-Inducing Activity,TIA) were detected in the peripheral blood of patients with advanced lung cancer. We studied the physical and chemical properties of factors. The purpose of this study is to test antibodies. Human lung cancer cell line (QG56), antiserum, antiserum The antibody was confirmed by quantitative ELIZA assay and TIA assay. There is a secretory substance in the supernatant of TIA cancer cell line. The molecular weight of TIA was determined by Western-Blotting method. The results showed that the molecular weight of TIA was 20-300000, and the molecular weight of TIA was 20-300000. The serum of fetal bovine fetus was not specific. The cancer cell lines of the present cancer cell line have no serum, and the cell lines of the cancer cell line are not serum-free. The animal animal is used to indicate the hyperactivity of cancer by using cancer transfer and TIA cancer transfer. The results showed that the patients with lung cancer were diagnosed as having sex with TIA, and the patients with lung cancer were diagnosed as having sex with each other. At present, the TIA government is in the process of moving the leadership of the government.
