项目摘要

The mRNA expression by reverse transcription-polymerase chain reaction (RT-PCR) method and immunohistochemistry of cytokine were studied with gingival biopsies from 20 cases of adult periodontitis and 4 cases of rapidly progressive periodontitis. Twenty-four cases were divided into two groups of mild type and severe type, which were defined by depth of periodontal pocket. The depth of periodontal pocket which is less than 6 mm is the feature of mild periodontitis. The severe periodontitis is characterized by the increased depth of periodontal pocket (more than 6 mm).Immunohistochemical study showed that immunocompetent cells in mild periodontitis reacted with interleukin 2,3 and 4. By contrast, interleukin 1alpha, 1beta, 2, 4 and 6 positive cells were frequently observed in severe periodontitis. By RT-PCR method, mRNA expression of interleukin 1beta in severe periodontitis was significantly higher than that of interleukin 1alpha in severe periodontitis.Expression of mRNA of interleukin 2, 3, 4, 5 and 6 was not detected in the inflamed gingiva.These results suggest that interleukin 1beta may play the important role of pathogenesis and chronic status.
应用逆转录-聚合酶链反应(RT-PCR)和免疫组化方法对20例成人牙周炎和4例快速进展性牙周炎牙龈组织中细胞因子mRNA的表达进行了研究。根据牙周袋深度将24例患者分为轻度和重度两组。牙周袋深度小于6 mm是轻度牙周炎的特征。免疫组化结果显示,轻度牙周炎牙周组织中免疫活性细胞与白细胞介素2、3、4呈阳性反应。而IL-1 α、IL-1 β、IL-2、IL-4和IL-6阳性细胞在重度牙周炎中多见。RT-PCR检测IL-1 β mRNA在重度牙周炎中的表达明显高于IL-1 α,而IL-2、3、4、5、6在炎症牙龈中均未检测到mRNA表达,提示IL-1 β可能在牙周炎的发病机制和慢性状态中起重要作用。
