The mechanism of the acquired resistance to the anti-proliferative effect of IL-1 on human melanoma cells.

人黑色素瘤细胞系经长期培养后，对人IL-1APHA的抗增殖作用产生抗药性。利用敏感和抗性克隆，我们研究了细胞获得抗性的机制。耐药细胞结构性地产生IL-1α。IL-1以自分泌的方式诱导耐药细胞产生IL-6，耐药细胞对IL-6的抗增殖作用也具有抵抗作用。在1个抗性克隆中，IL-6受体的mRNA水平很低，提示获得性抗性是由于缺乏IL-6受体所致。

A human melanoma cell line became resistant to the anti-proliferativr effect of human IL-1 apha after a long period of culture. Using the sensitive and resistant clones, we investigated the mechanism by which the cells acquired the resistance. Tesistant cells produced constitutively IL-1 alpha. The IL-1 induced IL-6 production in the resistant cells by an autocrine manner, and the resistant cells were also resistant to the anti-proliferative effect of IL-6. In one resistant clone, IL-6 receptor mRNA level was quite low, suggesting that the acquired resistance results from the lack of IL-6 receptor.
