Isolation and Molecular Phyogeny of the Most primitive Eukaryotes in Algae

1994年从枥木县汤本温泉那津的酸性温泉中分离到单细胞藻类、钙藻和硫藻。1995年在伊豆半岛的Atagawa、Mine和Shuzenji温泉等中性pH的温泉中发现了蓝绿藻（蓝藻）、聚藻球菌、磷藻和藻藻。然后，分别从caldarium、g.s uluraria和c.m merolae中提取总DNA，并采用PCR扩增其中的18个rRNA基因。结果表明，该基因主要来自于caldarium。红藻基因的部分DNA序列与其他红藻有很大的不同。我们推测红藻在系统发育上可能与标准的红藻相去甚远。

Unicellular algae, Cyanidium caldarium and Galdieria sulphuraria were isolated from a acid-hot springs, Nasu, Yumoto spa, Tochigi prefecture in 1994. Blue-green algae (Cyanobacteria), Synechococcus, Phormidium and Oscillatoria were found in hot-springs at neutral pH, such as Atagawa, Mine and Shuzenji spas, Izu peninsula in 1995.Then, total DNA were extracted from C.caldarium, G.sulphuraria and C.merolae and 18s rRNA genes of them were multiplied by using PCR.As a result, the gene was much obtained from C.caldarium. Partial DNA sequence of the gene of C.caldarium are considerably different from those of other red algae. We postulate that C.caldarium may be far from standard red algae phylogenetically.
