早期発症型歯周炎や難治性歯周炎に深く関与していると考えられているActinobacillus actnomycetemcomitansのポケット内における存在の有無及び治療による変動を検索することは重要であると思われる。そこで今回、A.actnomycetemcomitansの存在と血清型を同時に測定できることを目的として、ELISA法による簡易同定法の開発を試みた。まず、予備実験として血清型a、bおよびcのリファレンス菌株の培養液からペーパーポイントによりサンプリングしてきたA.actnomycetemcomitansにオートクレープ処理を行ない菌種および血清型特異性の認められる莢膜多糖抗原を抽出したのち、ELISA法による抗体価の測定を行なた。ELISA法は、ウサギに全菌体を用いて免疫して得られたポリクロナール抗体と本菌に対して抗体価が高かった患者抗血清の2つを用いてサンドイッチELISA法を行なった。その結果、A.actnomycetemcomitansに対して反応は認められるものの、血清型特異性が得られないということが判明した。そこで、用いる抗体を、血清型特異性が認められるオートクレープ抗原に対するモノクロナール抗体に、抗原を全菌体に変更した。しかし、現在所有しているモノクロナール抗体が血清型bのものしかないため、b型に関してのみ予備実験を行なったところ、血清型特異的にA.actinomycetemcomitansの抗体価が測定可能なことが判明した。そこで、歯周疾患者の約100部位の歯周ポケットからポケット内細菌のサンプルを採取し、サンドイッチELISA法を用いて測定を試みたところ、抗体価が認められたポケットは1つも存在しなかった。これは、日本人においては血清型bのA.actnomycetemcomitansはほとんど存在していないという他の研究者達の報告と一致した結果となった。しかし、これ以外の血清型に関しては不明のため、今後は血清型aとcの型特異莢膜多糖抗原に対するモノクロナール抗体を作製し、さらに検討を加えたいと考えている。

Early onset periodontitis and refractory periodontitis are closely related to the presence and absence of Actinobacillus actinomycetans and their treatment. A. actnomyetemcomitans and its serotypes were determined simultaneously by ELISA. A. actinomycetemcomitans culture medium was prepared for the identification of serotype a, b, and c capsular polysaccharide antigens. ELISA was used for the determination of antibodies. ELISA is used to detect the presence of antibodies against the whole cell and to detect the presence of antibodies against the whole cell. The results showed that A. actinomycetemcomitans was resistant to infection and serotype specificity was not detected. The antibody, serotype specificity, and antigen of the virus are all identified. The detection of antibodies against serotype b and serotype b specific A.actinomycetemcomitans is possible. About 100 sites of patients with dental diseases were collected and tested by ELISA. The presence of antibodies was identified. A.actnomycetemcomitans serotype b exists and is reported by other researchers. Serotype A and C specific capsular polysaccharide antigens are not identified.
