Two concepts of biomaterials are necessory for the maxillary reconstruction : one is to fill the bone defect by osteogenetic materials, and second is to keep the bone defect space for which the former material would be able to induce the osteoinductivity.The aim of this study is to investigate the os Resity of the experimental bone cavity and the interface between biomaterials and host bone using Expanded polytetrafloroethylene (EPTFE) Patch^<TM> composite with Bone morphogenetic protein (BMP) composite (EPTFE-BMP) membrane as a periosteum substitute. At the initial planning, fuman BMP was applied with this composite material, however, polification of it has noe been advanced.One mg of BMP extracted from bovine bone matrix was used for preliminary experimental study instead of fuman BMP.BMP was applied with the surface of the EPTFE Patches. Drill hole of 5mm in diameter was made with removal of surrounding periosteum in the femur of Japanese white rabbit and covered with EPTFE-BMP composite membrane. EPTFE alone was also used as a control on the other side. Two week and four week after implantation, all animals were sacrificed and the specimens were observed histologically. The results demonstrated that bone defect of the EPTFE-BMP composite membrane group was filled with new bone tissue mixed with bone marrow, and micro pores of the inner portion of the membrane were filled with new bone. Same new bone formation was also observed on the EPTFE alone group. The result demonstrated that there was no difference between EPTFE-BMP composite membrane and EPTFE alone group.
本研究采用生物材料-<TM>骨形态发生蛋白(BMP)复合材料(EPTFE-BMP)膜作为骨膜替代材料,观察了EPTFE-BMP膜对实验性骨腔的开放性及生物材料与宿主骨界面的影响。本研究采用从牛骨基质中提取的BMP替代Fuman BMP,并将BMP应用于EPTFE补片表面,进行了初步的实验研究。取日本白色兔股骨,去除周围骨膜,钻取直径为5 mm的孔,覆盖EPTFE-BMP复合膜。另一侧也使用单独的EPTFE作为对照。分别于植入后2周和4周处死动物,取标本行组织学观察。结果表明,EPTFE-BMP复合膜组骨缺损处填充有混合骨髓的新骨组织,膜内部微孔内填充有新骨。在EPTFE单独组中也观察到相同的新骨形成。结果表明,EPTFE-BMP复合膜与EPTFE单独组无差异。
