项目摘要

前立腺は,アンドロゲンによって活性化された尿生殖洞間充織から出されるパラクライン様物質が,その上皮に働いて誘導されるとの我々の仮説を検証するため,本年度は,アンドロゲン非存在下に,いろいろな増殖因子を尿生殖洞に与えてインビトロで培養し、アンドロゲンなしで誘導できるか否かを検討した。1.前立腺形成を誘発する増殖因子の探索胎齢16.5日雌・雄ラット胎児および胎齢15.5日雌・雄マウス胎児よりとり出した尿生殖洞を,アンドロゲン非存在下で,50ng/ml HGF,KGF,aFGF,bFGF,EGFなどの増殖因子の単独存在下でインビトロ培養した。増殖因子を加えなかった対照群では,いずれの場合も前立腺の発生はみられなかったが,調べた増殖因子のうち有効なものは,KGFおよびEGFであり,雌・雄にかかわらず前立腺芽の誘導が見られた。また,KGF+EGFでは,誘導効果は加算的であった。HGF,aFGF,bFGFは無効。2.前立腺とDNA合成細胞核の分布BrdLi法によりS期の核を検出したところ,誘導された前立腺芽中には,尿生殖洞上皮中に比し,多数のS期細胞が存在するのが確認された。無効の増殖因子を加えたものでは、上皮細胞の増殖は促進されても,前立腺芽の誘導はおこらないことがわかった。結論:本研究により,少くもEGFおよびKGFは,アンドロゲンの代りに,ラットおよびマウス雌・雄胎児尿生殖洞で,前立腺芽を誘導しうることが明らかにされた。

In the present year, we have investigated whether or not the growth factor in the urogenital cavity is induced in the absence of the hormone. 1. The female and male fetuses were cultured in the presence of 50ng/ml HGF,aFGF,bFGF and EGF alone. The growth factor is added to the female hormone group, and the female hormone is added to the male hormone group. KGF+EGF, induce. HGF,aFGF and bFGF are ineffective. 2. The distribution of DNA synthesis cell nuclei in the anterior gland was confirmed by BrdLi method. The S phase cells were detected in the anterior gland bud and in the urogenital cavity epithelium. The proliferation of epithelial cells is promoted by the addition of growth factors, and the induction of proglandular buds is promoted by the addition of growth factors. Conclusion: In this study, EGF and KGF were used to induce female and male reproductive cavity and anterior gland buds.
