Development of a novel detection method for abnormal prion protein

本研究的目的是建立一种新的检测异常朊病毒蛋白的方法。重组正常朊蛋白在大肠杆菌中表达。大肠杆菌中进行亲和层析纯化。通过用铜离子处理正常朊病毒蛋白制备变性朊病毒蛋白，其显示对蛋白酶消化的抗性。在本研究中，我们利用核酸适体和荧光试剂建立了一种高灵敏度和特异性的固相膜检测朊病毒蛋白的方法。

The aim of this research is to develop the novel detection method for the abnormal prion protein, which causes the prion disease. The recombinant normal prion protein was expressed in E. coli, and purified with affinity chromatography. The denatured prion protein, which indicates resistance against protease digestion, was prepared by the treatment of the normal prion protein with cupric ions. In this project, we developed highly sensitive and specific detection methods for the prion proteins on a solid phase membrane using nucleic aptamer and chemiluminescent reagents.
