Reproductive dysfunction observed in lactating cows has been attributed to metabolic adaptation which is needed to ensure energy demand for lactation. This is especially critical during the post-partum period of negative energy balance a time period where conception is highly desirable. The underlying mechanisms of reproductive dysfunction in dairy cows are incomplete understood. Between 6 and 30% of dairy cows develop a cystic ovarian follicle (COF) which is connected to anovulation, especially during the early post-partum period. Investigations on COFs are limited due to the lack of suitable models to induce and control COF formation. In the previous grant period we developed a procedure to reproducibility induce COF in dairy cows. We also identified and characterized several signal transduction pathways connecting metabolic stress due to oxidative stress with cellular alterations in the ovary of dairy cows. The aim of this grant proposal is to induce COF in dairy cows and to biochemically, endocrine and cell biologically characterize the resulting alterations on molecular as well as on cellular levels. A detailed knowledge of genesis and development of cystic ovarian follicles might offer a starting point to develop strategies in order to rescue this pathophysiological alteration and therefore improve reproductive fitness in dairy cows.
