The effect of antianesthetic drug on the GABAergic current induced by anesthetics were investigated.Method : Rats (17-20 days old) were used. Whole brain was removed and sliced in the cold icy saline. Thalalmic LD cells were identified under microscope. Whole cell recordings were made and GABAergic inhibitory currents were recorded while perfused with Ian solution bubbled with 95% O2 and 5% CO2. The effect of anesthetics (0.5% halothane and 0.5% sevoflurane) was first recorded, and then myristate or doxapram was applied with anesthetics for 10 to 15 min. and washed out.Result : Anesthetic-induced augmentation of GABAergic currents was slightly subsided by myristate or doxapram.
方法:选用17~20日龄大鼠,观察不同剂量抗麻醉药对GABA能电流的影响。取下整个大脑,放入冰冷的生理盐水中切片。显微镜下可见Thalalm LD细胞。灌流含95%O2和5%CO2的Ian液,记录GABA能抑制电流。记录麻醉药物(0.5%氟烷和0.5%七氟烷)的麻醉效果,然后加用肉豆蔻酸盐或多沙普兰10~15min。结果:肉豆蔻酸盐或多沙普兰对麻醉药引起的GABA能电流的增强有轻微的抑制作用。
