The basic and clinical study of effects of preoperative chemotherapy for the purpose of organ preservation surgery on oral squamous cell carcinoma

To elucidate the clinical significance of effects of preoperative chemotherapy on cell kinetics, we examined the changes in mean DNA content and mitotic index (MI) in oral squamous cell carcinoma, and attempted to correlate them with patients' survival rates. Nuclear DNA content and the MI in 118 specimens from patients before and after chemotherapy were measured using a cell image analyzer. It was suggested that image analysis of nuclear DNA content in combination with MI is useful for assessment of the effects of preoperative chemotherapy on cancer cell kinetics. This type of analysis may serve as a diagnostic aid for predicting patients' survival and selecting the suitable adjuvant therapy for individual patients with oral squamous cell carcinoma.One hundred twenty-two specimens from patients with oral squamous cell carcinoma were investigated by staining with a polyclonal antibody against pRb2/p130. pRb2/p130 expression was found to be correlated significantly with clinical stage, cervical lymph node metastasis, and tumor differentiation. Expression of pRb2/p130 may be a good prognostic indicator in patients with oral squamous cell carcinoma.
