The value of TSH receptor antibody will fall if thyroid function is normalized in Graves' disease. The duration of destruction is less than three months in destructive thyrotoxicosis. In order to explain these phenomenon, the hypothesis "thyroid hormone suppresses Th1 and stimulates Th2" was examined. Results ; (1) Whole blood was cultured with thyroid hormone (T3), and the amounts of IFN-gamma and IL-4 were measured with Invader RNA Assay. T3 slightly increased IFN-gamma and mildly increased IL-4. (2) Peripheral blood mononuclear cells from four people were cultured with T3, and the amounts of several cytokines were measured with TaqMan Cytokine Gene Expression Plate I. The results were different in every specimen. (3) Whole blood was cultured with PHA and T3, and the amount of IL-4 in a supernatant was measured by ELISA. IL-4 was not changed significantly by T3. (4) Peripheral blood mononuclear cells were cultured with PHA and T3, and the IFN-gamma or IL-4 producing cells were measured by ELISPOT. T3 slightly increased IFN-gamma producing cells, and slightly decreased IL-4 producing cells. Conclusions ; Thyroid hormone did not show a constant change in the Cytokine production from lymphocytes.
