Following investigation of several solvents and salts and the effect of pH to establish the new liquid-liquid extraction method using hydrophilic organic solvent, several pairs of the hydrophilic organic solvents and salts appeared suitable for the method. For liquid chromatography-mass spectrometry, ammonium acetate appeared most effective to salt out the drugs in the samples. These findings were then applied to identification and quantitation of benzodiazepines, barbiturates and antihypertensive medicines, such as barbital, phenobarbital, pentobarbital, secobarbital, nitrazepam, diazepam, bromovalerylurea, apronalide and prazosin. Furthermore, this new liquid-liquid extraction method was applied to the Triage test using gastric contents and serum specimens. The test results using samples extracted by the method was correlated better with the quantitative results by high performance liquid chromatography than those using unextracted samples.
