项目摘要

This study was designed to investigate the suppressive effect of HGF(hepatocyte growth factor) on GVHD(graft-versus-host disease) in nonmyeloablative preparative regimen bone marrow transplantation as the treatment of pancreatic cancer.In original plan, the effect of HGF on GVHD suppression was determined by dose reduction of immunosuppressant agents. But it was thought to unable to determine whether the reduction of immunosuppressant agents was possible by HGF because of the deficiency of T cell and B cell.So, we considered the laboratory model. We set the site of inplant of the cancer cell subcutaneous of the back of the SCID/B6 mouse. In this model, we determined the effect of HGF on GVHD and GVT effect.After plantation of pancreatic cancer cells, bone marrow and lymphocytes of spleen were transplanted. And we determined the changes in the size of implanted tumor and GVHD effects on small intestine and liver.Our data of GVT effect were scattering, and it was difficult to quantification of GVT effects. Although t is necessary to determine in more number of experimental model, it was confirmed that HGF may have possibilities to reduce the GVHD without reduction of GVT effect.
本研究旨在探讨肝细胞生长因子(HGF)对胰腺癌非清髓性预处理骨髓移植中移植物抗宿主病(GVHD)的抑制作用。但由于T细胞和B细胞的缺乏,无法确定HGF是否能降低免疫抑制剂的作用,因此我们考虑了实验室模型。我们将癌细胞的植入部位设置在SCID/B6小鼠背部皮下。在此模型中,我们观察了HGF对GVHD和GVT效应的影响。同时测定了移植瘤的大小变化及对小肠和肝脏的GVHD效应,GVT效应的数据比较分散,难以定量。虽然需要在更多的实验模型中确定,但已经证实HGF可能具有减少GVHD而不减少GVT效应的可能性。
