项目摘要

To develop a regenerative trachea of stable and safe mechanical strength, we attempted the making acellular tracheal scaffold and its application i.e., the tracheal reconstruction by using acellular tracheal scaffold.Dog or rabbit trachea enucleated were rinsed by Triton X-100 for 24 hours at ambient temperature. The component cells in trachea were removed almost all by Triton X-100 treatment. Rabbit and dog trachea were replaced by the acellular tracheal matrix lyophilized and sterilized. Acellular tracheal scaffold of rabbit kept the function of tracheal duct after grafting. Twenty-eight numbers of rabbit grafted survived for 14 to 60 days post surgery. However, all of rabbits grafted died by acellular trachea replaced was infected. Accelular tracheal scaffold of dog also kept the enough function of tracheal duct after grafting. Rabbit grafted acellular trachea did not show angiogenesis at grafted region enough but dog grafted accellular trachea obtained the results of good angiogenesis. The cellular infiltration and connective tissue proliferation in accellular trachea was not only found but also bronchial epithelium attracted on the lumen of acellular trachea grafted 2-months post grafting histochemically.Acellular tracheal scaffold may be an effective regenerative device for tracheal reconstruction.
为了研制出力学强度稳定、安全的再生气管,我们尝试了脱细胞气管支架的制备及其应用,脱细胞气管支架重建气管:犬或兔气管去核后,用Triton X-100室温冲洗24 h。TritonX-100处理后,气管内成分细胞几乎全部被去除。用冻干灭菌的脱细胞气管基质替代兔和犬气管。脱细胞兔气管支架移植后保持了气管导管的功能。28只移植兔存活14 - 60天。但脱细胞气管移植后的兔均发生感染而死亡。犬气管支架移植后仍能保持气管导管的功能。兔脱细胞气管移植后血管生成不足,而犬脱细胞气管移植后血管生成良好。脱细胞气管移植2个月后,组织化学观察发现脱细胞气管内有细胞浸润和结缔组织增生,并有支气管上皮细胞吸附于管腔上,脱细胞气管支架可能成为一种有效的气管重建再生装置。
