1. Developmental changes in maturation of immune related cells in the gut during the first 2 weeks post-hatch were determined in broiler chickens fed a corn-soybean meal based (CS) diet, semi-purified diet with high fat (HF) or high carbohydrate (HC). Expression of CD3 and IFN-y mRNA increased during 3 to 7 days of age in three sections of the gut determined. Further increase in CD3 was observed in mid and hind gut during 7 to 14 days. When chicks were fed the CS diet, expression of IL-2 mRNA in fore gut did not increased with age during the experimental periods, but the expression in mid section increased at 14 days of age and in the hind section at 7 days of age. Expression of iNOS mRNA was lowered at 3 days of age in the fore and mid gut but the expression was constant at the other days of age. The changes in iNOS expression in the hind gut were similar as that of CD3 mRNA. TLR-2 mRNA expression increased during 3 to 7 days of age in three section of the gut, while TLR-4 expression increased during 0 to 3 days of age. Chicks fed the HC diet showed higher IL-2 but lower IFN-y mRNA expression than birds fed the CS diet and had age related increase in IL-2 expression in the fore gut. Feeding the HF diet tended to decrease the expressions determined except TLR-2, regardless of age and section of the gut determined.2. The complete open reading frames of chicken TL1A contained 717 base pairs with an open leading frame, predicting a protein of 239 amino acids. ChTL1A mRNA expression changing by LPS stimulation was observed in most tissues, in particularly the highest levels were found in spleen and adrenal. Furthermore, recombinant ChTL1A has a dose-dependent cytotoxic activity to both L929 and chicken fibroblast.
