近年, 天疱瘡患者血中抗表皮細胞間抗体の抗原物質とデスモゾーム構成蛋白との関連が示唆されている. 本研究はデスモゾーム構成蛋白に対するマウスモノクローナル抗体を作成し, 天疱瘡抗体との関連を検討することを目的とした. デスモゾームはSkerrow & Matoltsyの方法に従い不連続蔗糖濃度匂配遠沈法を用いて, ウシ鼻表皮より分離精製した. この精製デスモゾームを免疫源としてBalb/cマウスを免疫し, 免疫マウス脾細胞とsp2/0細胞を用いて通常の方法によりhybridowaを作成した. ELISAを一次スクリーニング法として陽性wellを検索し, さらに, その陽性wellの中から蛍光抗体法により表皮細胞間に反応するwell1個を得た. 3回のlimitting dilutionによりクローニングし, さらに, プリスタン処理Balb/cマウス腹腔内に接種し, 腹水としてモノクローナル抗体を回収した. 蛍光抗体法により, この抗体は, 正常皮膚切片および培養細胞において, 表皮細胞間に強い反応性を示し, さらに, 細胞質にも若干の染色性を認めた. 精製デスモゾームを用いたWestern blot法により, この抗体は約240KDおよび約220KDの2本のバンドを染色した. 免疫電顕的に, この抗体は表皮細胞膜デスモゾームのattachment plaque部に反応した. 以上により, この抗体は, デスモプラキンIおよびIIに反応性を有する抗体と考えられた. 細胞質の反応性に関しては, 細胞質内にデスモプラキン蛋白の一部が存在する, 他の蛋白が同じエピトープを有する, 等の可能性が考えられるが, 今後, 検討が必要とされる. 各種天疱瘡患者血清とこの抗体によりブロッキング試験を施行したが, 天疱瘡抗体とこの抗デスモゾーム抗体の同一性は見出せなかった. 現在, 他のデスモゾーム構成蛋白に対するモノクローナル抗体の作成が進行中であり, これらの抗体を用いて, さらに, 各種天疱瘡抗体の抗原物質とデスモゾームとの関連を検討する予定である.

In recent years, the relationship between the antigenic substance and the component protein of anti-epidermal intercellular antibody in the blood of pemphigus patients has been shown. The aim of this study is to investigate the relationship between pemphigus and its constituent proteins. The method of Skerrow & Matoltsy is used to separate and purify the nasal epidermis without sucrose concentration and by remote precipitation. This is the first time that we've seen this. The ELISA method was used to detect the positive well, and the positive well was used to detect the positive well. 3. limiting dilution treatment Balb/c intraperitoneal inoculation, ascites and recovery of antibodies. Light antibody method: normal skin sections and cultured cells, strong reaction between epidermal cells, and some staining of cytoplasm. The purified protein was detected by Western blot. The antibody was about 240KD and about 220KD. The immune response of these antibodies to epidermal cell membranes is reflected in the attachment plaque. The above is the first time that the antibody has been tested. Cytoplasmic reactivity is related to the existence of a part of the protein in the cytoplasm, and the existence of other proteins in the cytoplasm. The identity of pemphigus antibodies and anti-pemphigus antibodies in sera from various pemphigus patients was investigated. Now, the preparation of antibodies against pemphigus is in progress, and the relationship between the antigenic substances of various pemphigus antibodies and the protein components is discussed.
