The genotype Blastocystis hominis is highly polymorphic. Therefore, a genetic marker would be a powerful tool for the identification or classification of B.hominis subtypes and could be used as a means to resolve the transmission route or origin of the parasite. To this end, approximately 100 Blastocystis isolates were collected from humans and animals, and these organisms were subjected to genotype analysis based on diagnostic PCR primers. A total of 9 PCR primers were developed from randomly amplified polymorphic DNA products of Blastocystis isolates recovered from humans or animals. Based on PCR amplification using diagnostic PCR primers, approximately 50 isolates recovered from humans were classified into four different subtypes. In addition, these PCR primers were examined with approximately 50 isolates recovered from animals and 3 isolates from chickens, 3 isolates from quails, and 2 isolates from monkeys were amplified with the diagnostic PCR primers. Therefore, these amplified strains were identified as zoonotic B.hominis strains.
