本年度は安定して遺伝子組み換え動物を供給できる環境を作ることに重きを置き、外部の施設で繁殖をしてくださっている先生と連携を行った。繁殖中の遺伝子組み換え動物の様子を確認しつつ、遺伝子組み換え動物以外の動物を用いながら、安定した手術手技の確立と、評価方法の確立を目指した。通常の動物でも侵襲を伴う手術であるため、毎回同じような神経損傷モデルを作製することに苦戦している。手術の麻酔方法もまだ吸入麻酔薬でしか行えておらず、静脈麻酔薬を用いての手術方法の実践や、顕微鏡で観察する際に静脈麻酔で維持させる装置の確立も並行して進めている。また、遺伝子組み換え動物でなくても本実験の目的を達成させるために同じ実験系を確立できないか試行錯誤をしており、代替となる蛍光色素などを検討している。脳神経細胞に蛍光カルシム蛋白を発現していない遺伝子組み換え動物を使用する場合には、観察可能な脳の領域も異なるため、本実験の目的を達成するのにふさわしい脳領域の観察が可能かどうか、手術方法も可能かどうかを含めて総合的に検討を行っている。動物実験を行うのにも限りがあるので、動物実験ができない期間には、実際の慢性疼痛患者の大規模コホート研究のデータを使用させていただきながら、実験データの処理方法にも関与する統計学の知識を学んだ。慢性疼痛患者の疫学的特徴や治療方法の実際を学ぶことで代替で観察する脳領域の候補についても理解を深めることができた。動物実験のデータをまとめていく上でも疫学統計の知識と技術は非常に重要であり、今後結果をまとめていく際に学んだ知識と技術を活かしたい。
