项目摘要

（１）忘却促進ペプチドの分泌細胞の同定忘却促進ペプチド遺伝子が発現している細胞を確認するために、GFPレポーター遺伝子用いて観察したところ、AWC神経を含む複数の頭部の神経細胞で発現が確認された。さらに、忘却促進ペプチドがどの細胞から分泌されることが必要なのかを確認するため、Cre-loxPシステムを用いてペプチド遺伝子の細胞特異的な発現阻害により行った。その結果、ペプチド遺伝子をAWC神経特異的に欠失した株では忘却が起きにくくなっていたことから、AWC神経からのペプチドが忘却に必要であることが示唆された。（２）忘却促進ペプチドの受容体が機能する細胞の同定忘却促進ペプチドの受容体遺伝子が発現している細胞を確認するために、GFPレポーター遺伝子用いて観察したところ、複数の頭部の神経細胞で発現が確認された。さらに、その結果と基に、受容体が機能するための細胞の同定を行うために、受容体変異体において細胞特異的に野生型遺伝子を発現させた表現型回復実験を行った。しかし、現在までに細胞の特定までには至っていないので、今後実験条件の検討などを行いながら、実験を継続していく。（３）忘却促進ペプチドが感覚記憶痕跡に与える影響の解析AWA神経にCa2+プローブを発現させたトランスジェニック株を用いて、条件付け前後・条件付け後の忘却の３段階で、AWA神経のジアセチルに対する応答を計測した。その結果、ペプチドとその受容体の変異体では条件付け後の回復後でもAWA神経に記憶痕跡が残っていたことから、忘却促進ペプチドはAWA神経にある感覚記憶痕跡に働きかけて忘却を促進していることが示唆された。
