In animals, formation of cell-matrix contacts is essential for embryogenesis and wound healing. Assembly and dynamics of such structures can be studied in tissue culture, and a large number of proteins have been found localized at the cytoplasmic face of these contact sites, where they are assumed to link microfilament ends to the plasma membrane. However, little is known on the regulation of cell-matrix adhesion sites, i.e. their assembly, maturation and disassembly in reaction to extra- and intracellular signals. In the project presented here, we would like to study the role of protein kinase C (PKC) in these events. Preliminary data describe the interactions of several isotypes of this kinase family with actin, vinculin and talin which are all essential components of cell-matrix contacts. We propose to study these interactions in detail and to expand these studies to other cell contact proteins. We will follow translocation of PKC isotypes during cell adhesion, spreading and migration. Colocalization of PKC with the contact proteins by immunofluorescence and by CLS microscopy of GFP-construct transfected cells will be combined with biochemical studies, using isolated recombinant proteins, cell fractionation, membrane-permeant crosslinkers and immuno-precipitation. Inhibitor and activator studies will reveal information on the role of PKC activity and PKC-dependent phosphorylation of contact proteins in these processes. Während der Embryogenese und der Wundheilung ist die Ausbildung von Kontakten zwischen Zellen und der extrazellulären Matrix ein Prozess von grundlegender Bedeutung. Aufbau und Dynamik solcher Kontakte, die morphologisch diskrete Strukturen darstellen, können in der Zellkultur untersucht werden. Auf der zytoplasmatischen Seite der vollständig ausgebildeten (reifen) Zell-Matrix-Kontakstellen stationärer Zellen wurde eine eindrucksvolle Zahl von Proteinen lokalisiert, die in vitro untereinander und mit Mikrofilamenten wechselwirken. Es wird daher angenommen, daß diese Proteine die membrannahen Enden der Mikrofilamente in Suprastrukturen organisieren, welche dann mit den Transmembran-Proteinen der Integrin-Familie verknüpft werden. Die Kinetik solcher Interaktionen, die Hierarchie der Komponenten sowie die Steuerung der Kontaktausbildung durch extra- und intrazelluläre Signale sind noch weitgehend unbekannt. Bei der Regulation dieser Vorgänge wird jedoch Proteinphosphorylierungs-Prozessen durch Kinasen wie der Proteinkinase C und der "focal adhesion kinase" eine große Bedeutung beigemessen.
