Cell-Matrix complexes or focal adhesions (FAs) are dynamic, membrane-associated anchoring structures that are essential for tissue formation in animals. They act as structural mediators between different signal transduction pathways and the actin cytoskeleton. Both, activity of individual protein components of FAs and their molecular interaction, as well as their role in transmitting external signals to the cytoskeleton depend on signals. In this project, we plan to study several FA components as examples of such functional interplay. To this end, these proteins will be purified as recombinant, full-length polypeptides and analysed with respect to their activity and complex formation as dependent on phosphorylation and phospholipid binding. Furthermore, using stable cell lines deficient for individual components, we will analyse various functional properties of such proteins and corresponding derivatives in microinjection studies. By combining data from both parts of this project, we hope to contribute to the understanding of the cellular significance of FAs as important communicators.
