Our research project consists of the following categories.(1) Construction of the experimental model of the dog with chronic and gastric fistulas which allows us continuous collection of pancreatic and gastric juice in the conscious state.(2) Establishment of the specific and sensitive radioimmunoassay for plasma cholecystokinin (CCK) that plays a physiologically important role for exocrine pancreatic secretion and gallbladder contraction.(3) Study on the effect of gastrointestinal hormones, such as Gastrin Releasing Peptide (GRP), Neuromedin C, Peptide YY (PYY), Peptide Histidine Isoleucine (PHI), on exocrine pancreatic secretion and plasma levels of gastrointestinal hormones including CCK to clarify the biological activity and physiological importance of these peptides.(4) Simultaneous measurment of exocrine pancreatic secretion and endogeneouslly released CCK on dogs with chronic and gastric fistulas following introduodenal instillation of fat or food stimulation to determine if CCK has true physiological importance.(5) Observation on the endogeneouslly released CCK in human and on the changes of the release of gastrointestinal hormones following resection of gut organs in the digestive surgery.(6) Comparative study of the immunohistochemical localization of the gastrointestinal hormone producing cells including CCK and radioimmunoassay measurement of tissue concentrations of gastrointestinal hormones.
