项目摘要

Flow cytometric DNA measurements were performed on 6 salivary gland tumors, consisted of 1 benign pleomorphic adenoma, 2 adenoid cystic carcinomas, 1 acinic cell carcinoma, 1 malignant salivary gland tumor and 1 adenocarcinoma. Non-neoplastic controls consisted of 12 differrent specimens obtained from submandibular salivary glands, lymphonodes, and oral mucosal epitheliums. DNA ploidy and cell cycle distributions were calculated from DNA histograms and compared to cytologic data.Only 2 tumors ( 1 malignant salivary tumor and 1 adenocarcinoma) were clearly diagnosed as malignancy from their cytologic smears. But in the other 3 malignant tumors, cytologic diagnosis was very difficalt due to lack of cytologic malignant characteristics.In the DNA analysis, 4 of the 6 tumors (1 benign pleomorphic adenoma, 2 adnoid cystic carcinomas and 1 acinic cell carcinoma) showed 'nonmal' DNA diploid histograms such as seen in non-neoplastic control tissues. The DNA histogram of malignant salivary tumor had a diploid pattern with a significant increase in the tetraploid peak. The specimen of adenocarcinoma contained two types of tumor cells : a DNA diploid peak and a distinct DNA aneuploid peak. This tumor seemed to be a multiclonal type. The percentage of cells in S-phase (S-index) for all the diploid tumors were lower than for non-neoplastic controls. This finding indicated that the proliferantive activity of these tumors were lower than normal tissues. The S-index for the aneuploid tumor was simillar to the mean value for the non-neoplastic tissues.DNA analysis can offer distinctive tumor biological properties such as multiclonality and proliferent activity, and may yield clinically valuable information.
