Immunohistochemical study on the rabbit cerebellum in early postnatal periods

Immunostaining patterns of the cerebellum toward a monoclonal antibody, 11-4G12 (MAb 4G12), produced against the adult rabbit olfactory bulb were analysed in baby rabbits and rabbit fetuses. In the early postnatal periods, MAb 4G12 labeled intensely Purkinje cells in the anterior lobe whereas the MAb labeled slightly Purkinje cells in the posterior lobe. The former belonged to a perisomatic dendrite stage whereas the latter did to a main dndrite stage. There are great differences in morphological characteristics and immunostaining intensities between the anterior and posterior lobes.The abovementioned regional differences in immunostaing intensity also were observed at embryonic day 25 in which Purkinje cells in both lobes have an immature morphology. Thus, it is concluded that the regional difference in immunoreaction to MAb 4G12 is not due to difference in Purkinje cell maturation. This suggests that Purkinje cells of the rabbit cerebellum during development are segregated by molecular heterogenuiety. From immunobloting analyses, an epitope against MAb 4G12 had estimated molecular weights of 26 kd units.
