Human plasma gelsolin was purified by ammonium sulfate precipitatio DEAE chromatography, Affi-Gel Blue chromatography and phenyl chromatography Antisera against plasma gelsolin were raised in rabbits by injecting the purified protein.A competitive enzyme immunoassay for the measurement of human plasm gelsolin was developed, using polystyrene beads as a solid phase and bovine interstinal alkaline phosphatase as a marker enzyme. Plasma gelsolin levels in normal persons ranged from 124 to 270 g/ml. In a patient with acute hepatitis, plasma gelsolin levels decreased during the presence of transaminase and bilirubin elevations. In patients with chronic hepatitis, the gelsolin level did not change markedly, including the period of acute exacerbation. In patients with hepatocellular cancer, the gelsolin levels elevated after transarterial embolization in comparison with pretreatment values.
