Xenopus laevisとX.borealisの2種のカエル胚を用い種々のキメラ胚作製もしくはパラバイオシスを行う事により, Tリンパ球幹細胞の胸腺原基への流入時期, 流入について幹細胞と胸腺原基との関連について調べた. (1)22時間胚で前部がX.1後部がX.bとなるキメラ胚を作り一定時間後心臓原基を除去して5日後の胸腺原基を調べ, 受精後3日9〜12時間に幹細胞が流入する事を確認した. (2)発生段階(stage)が12時間異なる2者の胚を用い, (1)と同様のキメラ胚を作製し5日後胸腺原基を調べてみると, 大半のキメラ胚で, 後部由来の細胞の胸腺原基への流入は認められなかった. このキメラ胚においては幹細胞と胸腺原基との間に12時間のagingの差があり, 幹細胞の流入にはそれ自身, もしくは胸腺原基のagingが関与している事が強く示唆された. この事を更に調査するために(3)stageの異なる(0〜24時間差)2者のカエル胚(young embryo,YE=X.laevis,old,OE=X.borealis)を用いパサバイオシスを行った. これによって流入現象と, 幹細胞と胸腺原基のagingとの関連が調査しうる. 同じstageの胚を用いたパラバイオシスでは受精後2日の時期に両者に血球の混在が認められ, 互いの血管系の連絡が確認された. 5日後の両者の胸腺原基には互いの細胞が存在し, この系において2者の細胞間の親和性の違いは無視しうると考えられた. この相互の流入はstageの差が3〜4時間のseriesでも認められた. しかし差が6時間のseriesになると, OE側胸腺原基にYE側の細胞は認められるが, YE側の胸腺原基へOE側の細胞は流入しない. この関係はstageの差が10, 13, 16, 24時間のseriesにおいても同様であった. この事は正常発生において受精後3日9〜12時間で胸腺原基へ幹細胞が流入しはじめるが, 6時間以内に流入が停止する可能性が示唆される. それは胸腺原基ではなく, 流入する幹細胞自身のagingに関与していると考えられる.
Xenopus laevis X.borealis two kinds of mouse embryos were used to conduct business operations, and the thymic primordia of T cells were inflowed during the period of inflow, and then flowed into the thymic primordia of the mouse cells.
