Die regulierte Exozytose von Neurotransmittern an Synapsen ist mit einer schnellen Regeneration synaptischer Vesikel durch Endozytose gekoppelt. Es ist anzunehmen, daß die Geschwindigkeit und Effizienz der einzelnen Transportschritte in einer hoch geordneten räumlichen Organisation der Maschinerien dieser Prozesse und/oder ihrer funktionellen Kopplung begründet liegt und daß hierbei cytoskelettäre Komponenten eine maßgebliche Rolle spielen. Daher soll die Präsynapsenorganisation und -funktion ausgehend vom Gehirn-spezifischen Protein Syndapin I näher untersucht werden. Syndapin I ist ein geeigneter Kandidat für derartige Studien, da dieses Protein sowohl in der Rezeptorvermittelten Endozytose als auch in der Modulation des Aktinzytoskeletts eine Rolle zu spielen scheint. Diese Studie soll daher diese Syndapin-Funktionen in neuronalem Kontext näher untersuchen. Darüber hinaus sind Untersuchungen zur Rolle von Syndapinen in der Gehirnentwicklung und -funktion geplant. Die so gewonnenen Erkenntnisse sollen zusammen mit generellen licht- und elektronenmikroskopischen Untersuchungen zur Synapsanorganisation und der Identifizierung neuer hieran beteiligter Proteine zu einem vertieften Verständnis der funktionellen und organisatorischen Zusammenhänge von den verschiedenen Membrantransportprozessen und der Modulation des Zytoskeletts in der Präsynapse liefern.
