Analysis of Bacterial Adherence Factors and Cystitis.

Type 1 fimbriated uropathogenic E. coli which inoculated into mouse bladder via the urethera did not adhere to the luminal surface of normal superficial cells. But the adhesions of type 1 fimbriated E. coli to the basal side of isolated superficial cells and to the isolated intermediate and basal cells were observed. These adhesions were inhibited by the addition of mannose. These observations suggested the possibility of bacterial adherence to bladder epithelium according to the removal of superficial cells in vivo. To remove the superficial cells from bladder epithelium, detargents, collagenase or trypsin were injected injected into the bladder via urethera. Trypsin was most effective for removals of superficial cells and did not damage the intermediate cells of bladder. Electron microscopy observation of type 1 fimbriated E. coli injected into trypsin treated bladder showed the adherence and colonization onto intermediate cells of bladder. These colonizations of type 1 fimbriated E. coli caused the experimental cystitis.
