Sensitivity test of anticancer agents using 31P NMP Spectroscopy

1)体外实验使用人胃癌细胞系，检测抗癌剂处理后磷脂的代谢。需要的癌细胞数量过多，31 P NMP谱峰不清晰。2）体内实验将人胃癌细胞株NU-GC-4接种于裸鼠皮下，第7天腹腔注射顺铂（10 mg、7.5mg、5 mg/Kg），第14天用~（31）P核磁共振法检测磷脂代谢。第21天测量肿瘤大小。将31 P核磁共振波谱结果与肿瘤大小进行比较。P肌酸和β ATP与肿瘤大小有较好的相关性。

1) in vitro ExperimentUsing human gastric cancer cell lines, the metabolism of phospholpids was examined after treatment of anticancer agents. Too much number of cancer cells were needed, and the pek of 31P NMP spectrum was not cleat. The sensitivity of anticancer agents was not clearly observed in this manner.2) in vivo ExperimentHuman gastric cancer cell line, NU-GC-4 was implanted in the subcutaneons space of nude mice.CDDP (10mg, 7.5mg, 5mg/Kg) was administered intraperotoneally at 7th day, and phospholipid metabolism was examined by 31P NMR Spectroscopy at 14th day. Tumor size was measured at 21 st day..The results of 31P NMR Spectroscopy were compared with the size of tumor. Pcreatine and beta ATP had a better correlation with the tumor size.
