Basic Research on Hormonal Treatment of Pancreatic Cancer

1.人胰腺组织中雌二醇（E_2）和雌激素受体（ER）的研究22例中有17例E2阳性细胞。几乎是阴性的。冰冻组织ER生化检测均为阴性.表皮生长因子、表皮生长因子受体和c-erbB-2癌基因蛋白在胰腺癌中的表达胰腺癌组织中EGF、EGFR和c-erbB-2癌蛋白的阳性率分别为72%、36%和28%。这三种蛋白检测阳性的患者的生存期明显短于检测阴性的患者。用银染核仁组成区（NORS）法研究人胰腺癌细胞的增殖能力。良性胰腺组织（正常胰腺组织1.86<plus-minus>士0.38，胰腺炎2.14士<plus-minus>0.53）与恶性<plus-minus>胰腺组织（4.02士1.19）Ag-NORs指数有显著性差异（P&lt;0.01）。胰腺癌Ag-NORs指数显著低于胃癌（6.35 ± <plus-minus>1.32）和结肠癌（7.66 ± 1.44）。本文研究了自发诱发胰腺炎和糖尿病的Wistar Bonn/ Kobori大鼠（WBN/Kob）的癌变过程。阿扎胞苷（50 mg/只，ip）在该大鼠中未诱导胰腺癌。接种7，12-二甲基苯并（a）蒽可诱导该大鼠和常规Wistar大鼠胰腺导管细胞样癌。

1. Studies on estradiol (E2) and estrogen receptor (ER) in human pancreatic tissues. E2-positive cells were immunohistochemically found in 17 of 22 cases. ER was almost negative. Biochemical assay of ER in frozen tissue was negative.2. Immunohistochemical expressions of epidermal growth factor (EGF), epidermal growth factor receptors and c-erbB-2 oncoprotein in human pancreatic cancer. The positive rates of EGF, EGFR and the c-erbB-2 oncoprotein in pancreatic cncer tissues were 72%, 36% and 28%, respectively. The survival period for patients who tested positive for these three proteins was statisticaly shorter than for those who tested negative.3. The proliferating potential of human pancreatic cancer cells were studied by Ag-Nucleolar Organizer Regions (NORS) staining. A significant difference in Ag-NORs index was found between benign (normal : 1.86<plus-minus>0.38, pancreatitis : 2.14<plus-minus>0.53) and malignant (4.02<plus-minus>1.19) tissues of pancreas(p<0.01). Ag-NORs index in pancreatic carcinoma showed significantly lower value than that in cancer of stomach (6.35<plus-minus>1.32) and colon (7.66<plus-mi4. The Wistar Bonn/ Kobori rat (WBN/Kob) that is induced pancreatitis and diabetes mellitus spontaneously was studied for the carcinogenesis. Azaserine (50 mg/body, ip) did not induce pancreatic cancer in this rat. Inoculation of 7, 12-dimethylbenz(a)anthracene induced the duct cell-like carcinoma in pancreas of this rat as well as conventional Wistar rat.
