イヌの骨髄の赤血球造血機能を知るために、正常な新生子犬-幼犬-成犬-老犬の各世代のイヌの骨髄について、赤血球系幹細胞(CFU-E)の分化・増殖性ならびにその存在量について骨髄培養による赤芽球コロニー形成法を用いて検討した。はじめに、骨髄細胞の基本的な至適培養条件について検討し、培養液としてはalpha-mediumを使用し、メチルセルロースを加えて半流動性培地とし、ウシ胎子血清が30%含まれる条件下で、1plate0.5mlのplate培養とし、細胞数を4×10^4個/plateとなるよう調整して、これに0.02-0.2Uのエリスロポエチン(EPO)を添加して、5%CO_2の条件下にて37℃48時間の培養を行う方法が確立された。以上の方法で骨髄細胞を培養した結果、CFU-Eの分化・増殖性については、各種年齢層のイヌの骨髄細胞はいずれもEPOの添加量に用量依存的に反応し、その反応性は同程度と考えられた。一方、骨髄細胞中のCHU-Eの存在量に関しては、コロニーの形成数において、年齢が若いものほどコロニー数は多く、加齢とともに数が減少する傾向にあった。なお、いずれの年齢層においても種差ならびに性差は認められなかった。また、各種疾患のイヌの骨髄について同様の検索を行ったところ、CFU-Eの存在量に関しては特に疾患に原因すると考えられるような傾向の差異は認められなかったが、増殖性の面では、腫瘍性疾患(抗癌剤使用により骨髄抑制のあるものを含む)症例、貧血症例、腎不全症例では若干の増殖性亢進の傾向が、また内分泌疾患や異栄養性疾患の症例においてはやや増殖性低下の傾向が示唆された。
