ティザ-(Ty)菌感染動物体の菌およびサイトカインの動態をRT-PCR法によって検索した。18匹のBALB/cAマウスにTy菌(MSK株)4.2×10^6を経口投与し,投与後1,3,5,7,14および21日目にそれぞれ3匹ずつから,肝臓、心臓,脾臓,盲腸および腸間膜リンパ節を採取し,RNA抽出後,Ty菌特異的または、オリゴ(dT)プライマーを用いてcDNAを合成し,得られたcDNAについて,Ty菌またはサイトカイン(IL-1alpha,GM-CSF,IFN-gamma,IL-6,TNF-beta,IL-4,IL-2およびIL-10)をPCR法によって検索した。菌投与後14および21日目については,血清を採取し,Ty菌抗体をELISA法によって検索した。Ty菌は,菌投与後3および5日目に盲腸および腸間膜リンパ節から,5および7日目に肝臓から,さらに14日目には心臓から検出された。サイトカインmRNAは,各臓器とも菌の出現に伴っておもにIFN-gammaおよびTNF-betaが検出された。脾臓からは菌は検出されなかったが,投与後3および5日目にIFN-gamma,1および7日目に1L-1alphaが検出された。その他の肝臓においては投与後7および21日目にIL-10,心臓においては3および7日目にIL-6が検出された。しかし各臓器とも,GM-CSF,IL-2およびIL-4は検出されず.また血清からの抗体も検出されなかった。今回の実験から,経口的に投与されたTy菌は盲腸から腸間膜リンパ節を経て肝臓そして心臓に至ことが観察された。また,Ty菌感染に伴い,各臓器においてはIFN-gammaおよびTNF-betaがおおきく変動することが示された。さらに,回復期の血清からは,抗体が検出されなかったことから,Ty菌感染防御には細胞性免疫の関与が示唆されたが,サイトカインの変動からは,免疫担当細胞の特定はできなかった。
