Inhibitory effects of an antiandrogen agent on experimental esophageal carcinoma in rats

The incidence of esophageal carcinoma (EC) in men are higher than those in women and the androgen receptor has been detected in human skin cytosol. The purpose of this study is to investigate an inhibitory effect of Casodex on EC in castrated rats (CR) and noncastrated rats (NR).The weight of the ventral prostate gland after administration of 'Casodex' mixed into daily solid meal for 10 weeks was measured. The weight of the ventral prostate gland in group of 'Casodex' 25 mg/kg (n=5,132+-30 mg) was significantly lower than that in control group (n=5,713+-133 mg) (p<0.001).The incidence of oesophageal cancer in rats given 'Casodex' for 10 weeks after administration of AMN for 10 weeks was examined histpathologically. The incidence of oesophageal cancer in noncastrated rats administered 'Casodex' as a dose of 25 mg/kg daily (n=7,71.4%) was not lower than that in noncastrated control group (n=9,77.7%). The incidence of oesophageal cancer in castrated rats administered 'Casodex' as a dose of 25 mg/kg daily (n=8,25.0%) was lower than that in castrated control group (n=11,45.4%). We considered that 'Casodex' as a dose of 25 mg/kg has an inhibitory effect on experimental oesophageal cancer in castrated rats.
