Sjogren症候群(SS)患者61例と健康成人20例の末梢血DNAについて、VκI IISub group共通のCDR1領域とJκ定常部領域をPrimerとしてPCRにて種々のCycleにて増幅し、共通のCDR2領域のProbeとしてSouthem blot解析を行った。その結果、30Cycle以上のPCRではSS・健常者共に高率に再構成bandが観察されたが、Cycle数を落とすと健常人では陽性例が減少した。25CycleのPCRではSS患者61例中53例(87%)に再構成bandを認めたが、健常人では20例中1例(5%)であった。以上より、SSではVκI II Sub groupのgermline遺伝子の再構成cloneが多く認めらめる事が明らかとなった。次に、再構成バンドをゲルより切り出し塩基配列を決定した。現在のところ、SSの1例から得られた1cloneの塩基配列しか得られていないが、結果はHumkv325/Humkv305遺伝子が使用されていた。今後は、更に多くの症例の多くのcloneより塩基配列を決定し、健常人において使用されている遺伝子との比較により、SSにおいてVκI IISub groupのどの遺伝子が多く使用されているか、更に、SS症例のうち悪性リンパ腫やリンパ増殖性病変を有する症例では、どの遺伝子が多く使用されているかを調べ、SSからのリンパ増殖性病変・悪性リンパ腫への進展する機構を解明したい。また、他の膠原病についても同様の検討を行い、VκI II Sub group遺伝子の高頻度の活性化がSSに特異的な現象であるのか、あるいは膠原病一般に見られる現象であるのかを検索する予定である。

Peripheral blood DNA from 61 patients with Sjogren's syndrome (SS) and 20 healthy adults was analyzed. The Cycle of PCR in the CDR1 field and the Jκ constant field common to the VκI IISub group was increased by Primer and PCR. Probe and Southern blot analysis in the common CDR2 field were performed. The PCR results above 30 cycles were higher than those of SS and healthy people, and the positive cases of SS and healthy people decreased. 53 (87%) of 61 patients with SS and 1 (5%) of 20 healthy individuals were found to have re-formed bands. The above, SS VκI II Sub group Second, the composition of the base is determined. Now, in the case of SS, Humkv325/Humkv305 is used instead of 1clone. In the future, there will be more cases of multiple cloning and gene allocation, healthy people will use more genes, SS will use more genes. The organization of SS is responsible for the development of SS. In addition, other collagen diseases are also studied. The high frequency of activation of VκI II subgroup genes is a specific phenomenon of SS. Collagen diseases are generally observed.
