胃癌・腸上皮化生において高頻度に遺伝し欠失を示す、目標遺伝子の単離、あるいは、遺伝子座の同定を目標として、以下のような実験を施行した。1 ゲノム・ライブラリーのスクリーニング大腸癌細胞株WiDrから作成したゲノムライブラリーをcripto cDNAにてスクリーニングしたところ、最終的にcw12及びcw3-1の2cloneが得られた。これらの部分的シークエンスを決定し、ホモロジー検索を行ったところ、cw12はγ-crystallinとの間に、相同性が認められたが、cw3-1は明らかな相同性を有する遺伝子は認められず、エクソン領域外の配列と考えられた。2 マイクロサテライト法による遺伝子異常の検索γ-crystallin遺伝子は2q23-24に位置することから、その近傍のマイクロサテライト・ローカスに関して、遺伝子異常を検討した。しかし、検討した24列の胃癌症例中、4例(17%)にRERを認めたのみであり、当初の報告に見られた、高頻度の遺伝子欠失は認められなかった。現在、以前にチトクロームp450との間に相同性を認めたクローンを用いて、ゲノム・ライブラリーの再スクリーニングを行っている。
