冠動脈バイパス手術例および先天性心疾患の根治術例から右心耳の断端を得、これをコラゲナーゼとプロテアーゼを用いた酵素法により単一心房筋細胞に分離した。これらの細胞を用いてwhole cell clamp法により急速に流入するNa電流(I_<Na>)を記録し、イソプロテレノールのI_<Na>に及ぼす影響を検討し、チャンネルへの作用を検討する事とした。I_<Na>を正確に記録するため、電極抵抗1メガオーム前後の大きな開口部を持つパッチ電極を使用し、実験浴槽内の液温を17°Cに保った。I_<Na>以外の電流を遮断するため、細胞外液にはCs^+,Co^<2+>,4-aminopyridineを、電極内液にはCs^+をそれぞれ添加した。また細胞外液のNa濃度は15mMと低濃度に抑え、電圧固定が確実となるようにした。保持電位を-140mVとし、-20mVへの脱分極パルスを与えた際のI_<Na>の最大値は、イソプロテレノール(1μM)を細胞外液に添加した前後で比較するとイソプロテレノール添加後に約10%の増加を認めた。次にI_<Na>の定常状態の不活性化曲線に及ぼすイソプロテレノールの影響を調べたところ、1μMのイソプロテレノール添加によりこの曲線は過分極側に移動した。これらの結果はモルモット、ウサギなどの哺乳動物で検討された他施設での結果と似ている。現在のところ実験を行った例数が未だ少なく、前述の様な変化に対する統計学的検討がなされていないため、今後の予定はこれらの実験例数を増やす予定であるとともに、イソプロテレノールのI_<Na>を修飾する機序がA-キナーゼを介するものであるかを検討するため、β受容体を介さず直接adenylate cyclaseを活性化するとされるforskolinおよび膜透過型のcyclic AMPであるdibutyryl cyclic AMPなどによるI_<Na>の変化を検討する予定である。
