まず第一段階として、成熟家兎に対して麻酔下で角膜から前房内に24ゲージテフロン針を刺入固定し、ヘパリン加生理食塩水をつないで水銀柱にて加圧し多結果、130mmHgに加圧するのが十分な虚血状態を得られる条件であることを確認した。さらに加圧持続時間を30分、60分、90分の3通りに設定して、その後30mmHgに圧をもどして、その間の網膜電位図(ERG)変化を記録した。ERGは反応の大きさのみではなく、振幅が最大になるまでの時間に相当する頂点潜時というパラメーターを考慮できるように、ERG波形がシャープになる明順応下の反応を測定した。その結果、130mmHg加圧すなわち虚血状態の時間が長いほど、虚血解除後のERG振幅及び頂点潜時の回復が遅延するという結果を得た。この結果をもとに、デキストロメトロファンをはじめとする抗虚血製剤を静注し、各虚血時間ごとのERG反応の回復速度の相違を確認すべく実験を施行した。現在までのところ、明順応下ERGに関しては有意な回復速度の差を得ていないが、暗順応下ERG反応に関しては差があるという結果が得られている。したがって今後は、明順応下と暗順応下の虚血に対する反応の相違に着目し、本法にさらに改良と検討を加えて実験を継続する。また、家兎眼に生理的食塩水のみの灌流下とデキストロメトルファン加生理食塩水灌流下の2通りで術中高眼圧状態を持続して硝子体手術を行い、術中、術翌日、術1週後のERGを測定し、その後眼球摘出して組織を検討した。まだ対象数が少ないため結果として発表できないが、今後継続して行う予定である。
