To determine whether Langerhans cell-free cultured human epidermal grafts survive for a prolonged period in allogeneic recipients, an experimental model using SCID mice reconstituted with human peripheral blood lymphocytes (hu-PBL) was established. In this model, hu-PBL were activated with anti-CD3 mAb in vitro and then transferred into SCID mice pretreated with anti-asialoGM1 Ab in vivo. Flow cytometry of the peripheral lymphoid organs demonstrated engraftment of hu-PBL (mainly T cells) 2-4 weeks after the transfer. Cultured human epidermal grafts were accepted by untreated by untreated SCID mice, when transplanted on the dorsal muscle and protected with a silicon transplantation chamber. Ten days after cultured human epidermal grafting on SCID mice reconstituted with allongeneic hu-PBL,however, detachment of the epidermis and infiltrantion of human CD3+ T cells in the graft bed were histologically confirmed. These results suggest that mice. This model will be usefull for analysis of the rejection mechanisms of cultured human epidermal allonrafts.
