1,PCR法を用いてABO血液型genotypeを決定した各ドナーの、vWF抗原量とvWF上のABO血液型抗原量を調べた。ELISA系で抗vWF抗体によって血漿中からトラップされるvWF抗原量と、そのvWFに対する血液型抗体(及びレクチン)の結合量に比例関係がみられたので、vWFあたりの血液型抗原量を指標として比較した。AA型(11例)とAO型(7例)を比較すると、抗A抗体の反応性はAA型の方が約25%強く、UEA-Iレクチンの反応性は逆にAO型の方が約35%強かった。また、vWF濃度は既報どおりAA型の方がAO型よりも高かった。一方、例数がまだ少ないが、亜型であるA2型の血漿vWFを調べると(3例)、1例でA型抗原量が少なく、H型抗原量の非常に多いものが見つかり、この検体では、vWF濃度も低かった。これらの結果から、同じ血液でもvWF上の血液型抗原量がgenotypeの影響を受けることが明らかになった。また、血液型抗原量(特にH型抗原量)とvWFの血中濃度に相関があること、すなわちO型でvWF濃度が低くなるように、genotype間でもH型抗原量の多いものほどvWF濃度が低くなる傾向があることが示唆された。2,vWFが血管内皮下組織のどのようなマトリクス成分と結合し、さらに結合することが引き金となってvWFが構造変化を起こし、血小板GPIbと相互作用できるようになるかどうかをELISA系で調べた。その結果、vWFは、III型コラーゲンと非常に良く結合するが、結合しただけではGPIb由来のグリコカリシンとは結合できず、蛇毒ボトロセチン存在下ではじめてグリコカリシン結合能を示すことがわかった。すなわち静水系では、マトリクス成分との結合のみではvWFの構造変化は起こらず、ずり応力やボトロセチン様の未知の補助因子がはたらく必要があることが示唆された(J.Biochemに発表)。
