1)各血液型単一ドナー血漿を電気泳動し、PVDF膜に転写後、抗A、抗B抗体、抗Hレクチンと反応する血漿成分をサーベイしたところ、二つの主要なタンパクが個々の血液型と一致して血液型抗原性を示した。アミノ末端配列分析、及び抗体との反応性から、これらはそれぞれフォン。ビルブラント因子(vWF)とalpha_2-マクログロブリン(alpha2M)であることが明らかとなった(Blood(1993)82,663-668)。2)単一血液型ドナーより約30種類の代表的な血漿タンパクを調整し、血液型抗体との反応性を詳しく調べたところ、vWF及びalpha2M以外のキャリアータンパク群、補体系タンパク、抗体、マトリスク成分に関してすべてABO血液型抗原性は認められなかった。しかし、癌患者血漿に含まれる癌胎児性抗原(CEA)に関して、血液型抗原を持つものが見いだされた。また、市販の第VIII因子(FVIII)濃縮製剤を調べたところ、FVRIIIも血液型抗原性を示した。3)免疫沈殿実験から、vWFとalpha2Mにおける血液型抗原量を調べたところ、vWFでは90%以上が血液型抗体により沈殿したのに対し、alpha2Mでは約5%が沈殿した。このことから、大部分のvWFに血液型抗原が存在するのに対し、alpha2Mでは一部の分子のみに血液型が存在することが明らかとなった。4)分泌型(Le^<a-b+>)、非分泌型(Le^<a+b->)各10検体の血漿中のvW量、血液型抗原量をELISAで比較したが、これらはいずれも各血液型ドナー間で有意な差を示さなかった。しかし、O型ドナーでは従来の報告通りvWF量は低い傾向を示した。以上、本研究でヒト血漿中のvWF以外に、alpha2M、FVIII、及び一部のCEAにもABO血液型抗原が存在することが新たに明らかとなった。これらの限定的な血漿糖タンパク質にのみABO血液型抗原が存在することは、これらの産生が、大部分の血漿糖タンパク質の生合成経路(産生部位)とは異なる部位で起こることを示唆し、今後産生部位の特定を試みる予定である。また、FVIII濃縮製剤もまた血液型抗原を含むことが明らかとなったことは、今後血液型別に血漿製剤を調整することでさらにその血中寿命が改善される可能性を示唆するものである。
